It is generally recommended to wait at least 60 days after entry on B2 visa to marry a U.S. citizen.

After marriage, all sponsorship paperwork can be filed concurrently. Your spouse will be able to continue to remain in the U.S. until she receives LPR (lawful permanent resident status) and thus you will not be separated from your spouse during the sponsorship process.
