There's a property in which my mother & my uncle have equal share. We are 3 daughters & 1 son to our mother & have given our share to our mother. Can my mother create a Will in which she can mention that after her death, her share of property should go to her son only with which we daughters are absolutely fine...

Yes, your mother can write up her Will of her 50% share in the property in the name of your brother.
