499/500 IPC is criminal action for which the accused would be imprisoned and you may be suitable compensated. Civil Defamation suit may be filed separately for damages as per law of torts for which you have to pay heavy court fees (approx 1% varies from state to state). You may file the civil suit now or after the accused is convicted in the criminal case. In case the defamation is filed now, it will run parallel with the criminal case and you don't run the risk of non receipt of damages even if the accused is acquitted, meaning thereby that if you dont file a civil suit now and the accused gets acquitted (after the limitation for civil defamation ends, as the trial will take 5-6 years) you cannot file for damages in case of his or her acquittal.
