Adminstration of a trust

Can a father(who has a sizable estate) bypass a son and leave everything to the grandchildren when creating a trust?

Yes. A person can leave their estate to whomever they wish (with certain provisions for a surviving spouse). Children can be disinherited. However, if a sizable estate is being left to grandchildren, the impact of possible generation-skipping transfer tax (in addition to estate tax) should be considered.
