My husband's cousin is a Canadian citizen who would like to work and live in New York, USA. What would he need to do to start the process?

He should first apply for jobs and see whether any employer is willing to hire him and apply for a work visa on his behalf. Depending on his academic background and the job offered, he should consult an immigration attorney and see whether he qualifies for a TN or H-1B visa.
