I typed the manuscript for a book by a gentleman who died before getting a copyright. Is it possible for his spouse to copyright and publish?

Yes, the spouse can claim the copyrights. You cannot claim the copyrights if you were merely the typist, or if you were paid for your services ("work for hire"). The copyrights pass into the deceased's estate, and his wife would be the first to claim on the estate.
